Four substances made by the pregnancy are measured in the mother's blood, usually between the fourteenth and twentieth week. Those levels are read together with your age, weight and the exact stage of pregnancy taken from the scan, and the laboratory works out a statistical risk. It is a screening test, so what comes back is a probability rather than a diagnosis.

Doctors order it for routine antenatal screening in the second trimester, particularly when the first-trimester scan and blood test were not done in time.
