Sputum is the thick material brought up from the lungs, not saliva from the mouth. It is smeared on a slide and stained with a dye that clings to the tough outer coat of the tuberculosis germ, which is why these organisms are called acid-fast bacilli. The slide is then examined under a microscope.

Doctors order it for a cough lasting more than two weeks, blood in the sputum, evening fever with weight loss, and to follow the progress of tuberculosis treatment.
