Adult blood contains mainly one type of haemoglobin, with small and fairly fixed amounts of two others. The sample is passed through a system that sorts these types apart, so abnormal forms such as those of thalassaemia or sickle cell disease can be picked out and the share of each measured.

Doctors order it for anaemia that has not improved with iron, a family history of thalassaemia or sickle cell, and screening before marriage or pregnancy.
